学术成就

长期从事细胞命运调控的表观遗传机制研究,主要的研究方向(1)胚胎干细胞干性维持以及向生殖祖细胞定向分化的表观机制研究;(2)肿瘤细胞的端粒染色体的表观遗传维持机制。应用生物化学技术剖析组蛋白去甲基化酶PHF8的作用机理以及小鼠干细胞定向分化成神经细胞中的重要生物功能。该研究结果为XLMR疾病提供了表观遗传学的发病机理(Cell Research,2010,共同第一作者排二);首次探究比较分析了同一去甲基化酶家族蛋白PHF2和PHF8在rDNA转录上的不同作用(Journal of Biological Chemistry,2014,共同第一作者排一);筛选和鉴定了HP1BP3蛋白在染色质结构调控中的作用机制(Nucleic Acids Research,2020,共同第一作者排一);以及揭示了类泛素化(SUMO)修饰抑制整体基因转录的新机制(Cell Research,2018,共同第一作者排二)。通过多能性因子(Oct4和Sox2)的蛋白组筛选中发现了维持干细胞多能性的新因子Ddx56蛋白(Stem Cell Research & Therapy,2020,独立通讯);还发现间充质干细胞在皮肤损伤修复中新的应用策略(Stem Cell Research & Therapy,2020(a-b)(2021),共同通讯)。此外,总结了重复序列如逆转录转座子在多能性维持中的重要作用(Cell Regeneration,2020,共同通讯);以第二或三发明人获得国家专利2项:(1)Bend5蛋白在提高胚胎干细胞诱导分化成原始生殖祖样细胞方面的应用(第三发明人)。(2)端粒结合蛋白HP1BP3在制备肿瘤细胞调控剂中的应用(第二发明人)。目前为止,本人在Cell Research、Nucleic Acids Research、Journal of Biological Chemistry等刊物上发表论文19篇,其中第一作者或通讯作者(含共同)11篇。获得国家自然科学基金青年项目、面上项目和广东省自然科学基金项目资助。
