研究方向

病原细菌/人体微生物组与宿主互作; 益生菌资源挖掘及其改善疾病的分子机制

         微生物与人类宿主之间的相互作用是生物物种间最复杂的相互作用之一。一方面,在与人类宿主亿万年共同进化的过程中,共栖微生物群(Commensal)形成了独特的微生态系统,成功适应了宿主环境。另一方面,外源性的病原菌(Pathogens)或共栖有害微生物则能突破宿主的免疫屏障,实现感染。肠道菌群是肠道内寄居的种类繁多的微生物的总称。鉴于人体胃肠道微生物群细胞的总数是人体细胞总数的数倍,编码的基因数量可达人体自身基因数量的150倍,因此被称为“人体第二基因组”和“新器官”。人体胃肠道微生态系统是人体代谢的重要参与者,代谢产生的短链脂肪酸等促进肠道上皮细胞生长与分化;肠道菌群之间可以通过如分泌系统分泌的效应蛋白等维持菌群的平衡。肠道菌群平衡的失调(Dysbiosis)与约90%的人类疾病密切相关,包括胃肠道疾病、炎症、代谢障碍类疾病(如肥胖、糖尿病等)、心血管疾病、免疫疾病、癌症和精神性疾病等。对肠道微生物组与疾病关系的研究,将为开发基于微生物的疾病诊断、疾病干预、治疗和益生菌产品的研发等奠定基础。目前该领域的研究正从微生物与宿主互作的“关联性”向“因果性”转变。本实验室围绕微生物组(Microbiome,包括微生物及其代谢产物及肠道自身代谢物)与宿主相互作用的机制,开展如下研究:

(1)肠道微生物组与宿主的互作及其分子机制:我们综合运用基因组学、代谢组学、基因组操作和生物信息学等技术,研究肠道微生物组或肠道细菌与炎症、肥胖症和癌症等的发生、发展的关联性及其分子机制;发现生物标志物(biomarker)及可成药靶点(druggable targets)。

(2)肠道微生物培养组学及益生菌资源挖掘与应用: 运用培养组学策略分离纯化各类肠道益生菌,并评估其临床应用价值;开展临床干预实验,研究其益生的分子机制。

(3)肠道微生物的基因组遗传操作:为治疗性工程化微生物活菌药物设计及肠菌的精准编辑奠定基础。

(4)靶向抗菌药物筛选及作用机理:以细菌分泌的效应蛋白作为靶位点,筛选活性先导物,获得避免耐药性产生的新型抗生素:结核分枝杆菌(Mycobacterium tuberculosis, Mtb)是结核病的病原,感染了我国近三分之一的人口,是国人健康的重大威胁,急需新的抗结核药物。我们以结核分枝杆菌关键效应蛋白为靶点,建立相应的筛选系统,从微生物代谢产物中筛选效应蛋白的抑制剂,并研究其作用机制,期望获得新型的、能避免耐药性产生的抗结核药物。
